Quick answer: SAMe & TMG is a lemon-flavored powder from Xymogen that puts two methyl donors in one stick pack: 400 mg of S-adenosyl-L-methionine (SAMe) and 600 mg of betaine anhydrous (TMG, trimethylglycine). The label dose is one stick pack a day, taken 1–2 hours before or after a meal, either dissolved on the tongue or stirred into 2–4 oz of water. It suits adults whose practitioner wants SAMe for mood, liver and joint support and likes the idea of a methylation backup in the same serving.
| Form | Lemon-flavored powder in single-serving stick packs (about 2.6 g each) |
| Per stick pack | 400 mg SAMe (as the 1,4-butanedisulfonate salt), 600 mg betaine anhydrous (TMG), 75 mg calcium; 5 calories, 1 g carbohydrate |
| Servings per box | 30 (a one-month supply at the label dose) |
| Who it suits | Adults wanting practitioner-guided methylation, mood, liver and joint support who would rather not swallow capsules |
| Take it | Once a day, 1–2 hours away from meals |

Gwen Halvorsen has adjusted insurance claims for twenty-six years, and she reads a supplement label the way she reads a contractor’s estimate: line by line, hunting for padding. At 52 she had a SAMe capsule bottle and a separate jar of TMG tablets on the counter, both suggested by her practitioner, and she distrusted the arrangement on principle. Two bottles meant two refills, two ways to forget. Then she pulled up the back panel of a Xymogen box and found both numbers on one label, in one lemon stick pack, and did the arithmetic before she did anything else.
Here’s what that label actually says, and how to decide whether the combination is worth it for you.
Xymogen® SAMe & TMG is a once-a-day powder that pairs a full 400 mg serving of SAMe with 600 mg of trimethylglycine. Xymogen describes it as support for biochemical reactions that require methyl groups, neurotransmitter synthesis and a healthy mood, the conversion of homocysteine toward glutathione, liver function and joint comfort. It comes as 30 single-serving stick packs, so there’s no scoop and no bottle to keep sealed between doses.
What is in SAMe & TMG, and what does each ingredient do?
Each stick pack holds two active ingredients, SAMe and TMG, plus a small amount of calcium that comes from the salts used to keep the SAMe stable. Here’s the label, ingredient by ingredient.
| Ingredient | Amount per stick pack | What it does |
|---|---|---|
| S-adenosyl-L-methionine (SAMe) | 400 mg, as SAMe 1,4-butanedisulfonate | The body’s main methyl donor; feeds methylation and the transsulfuration pathway that leads to glutathione |
| Betaine anhydrous (TMG) | 600 mg | Donates a methyl group to homocysteine, turning it back into methionine, the raw material for new SAMe |
| Calcium | 75 mg (6% DV), as carbonate, oxide and chloride | Mostly a by-product of the stabilizing salts; a small contribution to daily intake |
| Other ingredients | — | Xylitol, citric acid, malic acid, stearic acid, silica, natural lemon flavor, turmeric extract (for color) |
SAMe: 400 mg of the body’s main methyl donor
Your cells make SAMe from the amino acid methionine and ATP, then spend it as a methyl donor in reactions that build neurotransmitters, phospholipids for cell membranes, proteins and DNA. A review by Teodoro Bottiglieri in the American Journal of Clinical Nutrition describes SAMe as the precursor to three pathways at once: methylation, aminopropylation (which makes polyamines) and transsulfuration, the route that ends in cysteine and glutathione. That last pathway is why Xymogen ties the formula to liver function and antioxidant support, and why one compound gets credited with mood, liver and joint roles.
TMG: 600 mg of betaine for homocysteine recycling
Once SAMe donates its methyl group, what’s left eventually becomes homocysteine. Your body has two ways to turn homocysteine back into methionine: one runs on folate and vitamin B12, the other runs on betaine. TMG feeds the second route. In a Wageningen trial in men and women with mildly elevated homocysteine, 6 g of betaine a day for six weeks lowered fasting homocysteine by 1.8 µmol/L compared with placebo, and it blunted the rise in homocysteine after a protein-style methionine load.
Be clear-eyed about the dose, though. A 2013 meta-analysis of five randomized trials in healthy adults found that at least 4 g of betaine a day for at least six weeks lowered plasma homocysteine, by about 1.2 µmol/L on average. The 600 mg in each stick pack is a fraction of that, so think of TMG here as a supporting methyl donor that complements the SAMe, not as a stand-alone homocysteine dose.
Why combine SAMe and TMG in one stick pack?
Because they sit on opposite ends of the same cycle: SAMe hands out methyl groups, and TMG helps recycle the homocysteine left behind back into methionine. Putting both in one serving gives a practitioner a single product that supplies the methyl donor and supports the recycling step, which is the logic behind the pairing on the label.
There’s also a practical reason, and it’s the one Gwen cared about. Two bottles means two refill dates and two things to forget. The powder format is a second practical point: anyone who struggles with capsules can let it dissolve on the tongue. The trade-off: you can’t adjust the SAMe and TMG doses separately when they’re sealed in the same pack.
If your practitioner is also thinking about the folate and B12 side of the cycle, that’s a separate conversation. Our guide to Xymogen Methyl Protect covers one of Xymogen’s B-vitamin methylation formulas.
How do you take SAMe & TMG?
Take one stick pack a day, 1–2 hours before or after a meal, or as your healthcare practitioner directs. Xymogen gives two ways to take it: pour a small amount into your mouth and let it dissolve, repeating until the pack is empty, or stir the whole pack into 2–4 oz of water or another liquid and drink it within 15 minutes.
| Question | What the label and research say |
|---|---|
| Dose | 1 stick pack daily (400 mg SAMe + 600 mg TMG) |
| When | 1–2 hours before or after a meal. The label doesn’t specify morning or evening; pick a time you’ll keep |
| With or without food | Without. Use water, not a smoothie or a meal replacement |
| How to mix | Dissolve on the tongue a little at a time, or stir into 2–4 oz of liquid and drink within 15 minutes |
| How long before you judge it | The label sets no timeline. Agree on a check-in date with your practitioner before you start |
| Upper limit | No Daily Value is set for SAMe or TMG. Stay at one pack a day unless your practitioner directs otherwise |
- Stick to water. The 15-minute window on the label suggests the powder shouldn’t sit mixed for long, so mix it right before you drink it.
- Count your other SAMe. If you already take a SAMe capsule, SAMe & TMG replaces it rather than adding to it. One stick pack equals two 200 mg capsules.
- Don’t tear into a damaged pack. The label says not to use a stick pack that’s damaged.
- Store it cool and dry, and well out of reach of dogs. The xylitol that sweetens it is harmful to pets.
Does oral SAMe actually get absorbed?
Some of it does, but less than you might expect, which is one reason a 400 mg serving is common. In a pharmacokinetic study of healthy volunteers given 1,000 mg of SAMe as enteric-coated tablets, blood exposure after an oral dose was only about 2 to 2.6% of the exposure from the same dose given by IV, and levels peaked around five hours after the dose. The tablets were well tolerated, and SAMe didn’t build up in the blood over five days of dosing.
Two honest caveats. That study used a different SAMe salt and a coated tablet, not a lemon powder, and no one has published absorption data on this stick pack itself. And SAMe is also made inside your cells all day long, so blood levels are only part of the picture. What the research does support is following the label: take it away from food, and don’t assume more is better.
Who is SAMe & TMG for, and who should skip it?
It’s best for adults whose practitioner wants a full 400 mg serving of SAMe and sees value in adding betaine for methylation support. That usually looks like:
- Adults looking for nutritional support for a healthy mood and positive outlook
- People who want support for normal liver function and glutathione production
- Adults looking for help maintaining joint comfort
- Anyone whose practitioner is focused on methylation and normal homocysteine metabolism
- People who find capsules hard to swallow, or who travel and like single-serving packs
Skip it, or talk to your provider first, if you:
- Have bipolar disorder. Xymogen’s label calls for special caution, and the NIH National Center for Complementary and Integrative Health (NCCIH) notes SAMe may worsen mania.
- Take an antidepressant or another serotonin-raising product, such as L-tryptophan or St. John’s wort. NCCIH lists these as possible interactions; your prescriber needs to know before you add SAMe.
- Take levodopa. NCCIH notes SAMe may reduce its effect.
- Are pregnant, nursing or trying to conceive. The label asks you to check with your practitioner, and NCCIH says SAMe’s safety in pregnancy hasn’t been established.
- Have a weakened immune system. NCCIH flags a theoretical concern for people who are immunocompromised.
- Have a dog in the house and a habit of leaving packs on the counter. Xylitol is harmful to pets.
Side effects are uncommon and usually minor. NCCIH lists nausea and digestive upset as the most likely ones. Not sure which list you’re on? Holistic Health Partners offers a free product consultation, and it’s worth bringing your medication list.
How does SAMe & TMG compare with other SAMe supplements we carry?
It’s the only one of these that adds TMG and the only powder; the capsules give you more control over the SAMe dose, and one of them costs less per milligram of SAMe. Prices below are Covenant Health Products list prices as of November 2026.

| Best for | One step: SAMe plus a methylation backup, no capsules | A lower 200 mg daily serving | Lowest cost per mg of SAMe; flexible 200–400 mg | SAMe paired with active folate |
Here’s the honest read. If your practitioner only wants SAMe, Pure Encapsulations® SAMe is the less expensive way to get it: a 60-capsule bottle supplies 12,000 mg of SAMe, the same total as a box of SAMe & TMG, and lets you take 200 or 400 mg a day. If a lower dose is the plan, Xymogen SAM-e capsules give you 200 mg a day, each sealed in its own blister.
If your practitioner prefers to pair SAMe with methylated folate rather than betaine, Ortho Molecular Products® makes a SAMe formula with Quatrefolic, available through our clinic after a free appointment.
SAMe & TMG earns its price premium when you want the full 400 mg of SAMe and the betaine together, in a format that doesn’t involve swallowing anything.
Is SAMe & TMG worth it?
Yes, if you and your practitioner want a full 400 mg serving of SAMe with betaine on board and you’d rather drink or dissolve it than swallow capsules. Xymogen SAMe & TMG gives you both methyl donors in one lemon stick pack, a clear label, and a one-month box.
It’s the wrong pick if you have bipolar disorder or take a serotonin-affecting medication without your prescriber’s OK, if you only need SAMe and want the lowest cost per milligram, or if you were hoping the 600 mg of TMG would match the multi-gram doses used in homocysteine research. It won’t.
Gwen, for what it’s worth, kept her receipt from the switch. She says the math worked out. She also says she’ll be the judge of everything else.
